اسکلروستین
اسکلروستین (انگلیسی: Sclerostin) پروتئینی است که در انسان توسط ژن «SOST» کُد میشود[4][5] که بر روی بازوی بلند کروموزوم ۱۷ واقع است.[6]

این مولکول، یک گلیکوپروتئین است که توسط سلولهای استخوانی (اوستئوسیتها) ترشح میشود و خواص ضدآنابولیک در جریان استخوانسازی دارد[7] و با مهار «مسیر سیگنالدهی Wnt»،[8][9] موجب کاهش ساخت استخوان میشود.[8]
این پروتئین علاوه بر اوستئوسیتها، در برخی کندروسیتها هم بیان میشود و استخوانسازی را توسط سلولهای استخوانساز (استوئوبلاستها) مهار میکند.[10][11][12]
تولید اسکلروستین، توسط هورمون پاراتیروئید،[12][13] فشارهای مکانیکی بر روی استخوان[14] (مثلاً ورزش) و برخی سیتوکینها نظیر «پروستاگلاندین E2»،[15] «اونکوستاتین M»، «کاردیوتروپین-۱» و «فاکتور مهارکنندهٔ لوکمی» مهار میشود.[16] بالعکس، تولید اسکلروستین توسط کلسیتونین تحریک و افزایش مییابد.[17] در نتیجه، خودتنظیمیِ فعالیتِ سلولهای استخوانساز (استوئوبلاستها) توسط یک سیستم فیدبک منفی کنترل میشود.[18]
جهش در ژن تولیدکنندهٔ این پروتئین منجر به بروز «بیماری فان بوخم» و اسکلروستئوز میشود.[6]
